Cagrilintide, the C20-acylated amylin analogue, in a 5 mg freeze-dried fill
This is the smaller of two cagrilintide fills: 5 mg of the long-acting amylin analogue as freeze-dried powder, half the 10 mg main listing. Cagrilintide keeps the 37-residue amylin backbone with its disulfide and amidated end, adds prolines that limit fibril formation and carries a C20 diacid for albumin binding. What the research covers
Human amylin forms amyloid fibrils, and a 1990 study placed the cause in residues 20 to 29, where a single proline at position 28 almost completely stopped synthetic peptides from forming fibrils1. Cagrilintide has prolines at 25, 28 and 29. Kruse and colleagues described the result as a stable, lipidated analogue with a long duration of action, developed with obesity as the indication2. Frequently asked questions
Is the 5 mg vial the same peptide as the 10 mg?
Yes: the same sequence, disulfide and C20 side chain, with a theoretical mass of 4409.0 Da. It is a separate lot with its own certificate number. In moles, 5 mg is about 1.1 µmol, a little less after allowing for residual water.
What concentrations does a 5 mg vial give?
That depends only on the diluent volume: 5 mg/mL in 1 mL of bacteriostatic water, 2.5 mg/mL in 2 mL. The reconstitution table on this page lists more volumes, and the peptide calculator handles any other.
