Retatrutide is an acylated peptide agonist of the glucagon, GIP and GLP-1 receptors, developed under the code LY3437943 and still investigational. The 50 mg vial is the second-largest fill, lyophilized and tested as its own lot. Behind it lies an older idea, a single peptide aimed at three metabolic hormone receptors, which was tested in rodents years before the retatrutide trials.

What the research covers
In 2015 Finan and colleagues described a monomeric peptide triagonist with matched activity at the GLP-1, GIP and glucagon receptors and no cross-reactivity at related receptors1. In rodent obesity models it outperformed the dual co-agonists and single-receptor agonists it was compared with1. Retatrutide applies the same three-receptor principle with a different balance: its discovery paper reported more activity at the GIP receptor than at the glucagon and GLP-1 receptors2. How each lot is tested
Every lot is tested in PrymaLab’s own laboratory in Ashland, Kentucky before release: RP-HPLC for purity, LC-MS for identity, Karl Fischer titration for water, LAL for endotoxin and USP <71> for sterility. Frequently asked questions
What separates the 50 mg vial from the 10 mg main fill?
The quantity of powder and nothing else: sequence, side chain and theoretical mass are identical. 50 mg is roughly 10.6 µmol, five times the main fill, and each 50 mg lot has its own certificate.
How long does reconstituted retatrutide stay stable?
None of the papers cited here measured it. The safety data sheet says to keep solutions at 2 to 8 °C, protected from light, and to use them within a period your own laboratory has established.
