This vial holds 15 mg of semaglutide as freeze-dried powder, the middle of the five fills. Semaglutide is a GLP-1 receptor agonist made of 31 amino acids plus a side chain on lysine 26, built from a γ-glutamic acid, two OEG spacers and an 18-carbon diacid. Much of the chemistry literature on semaglutide is about that side chain, which binds serum albumin, and the clinical programmes came after it.

What the research covers
The side chain was the main design variable. Lau and colleagues reported that the fatty acid and the way it was linked to the peptide governed albumin affinity, receptor potency and the duration of exposure1. After intravenous delivery to mini-pigs, plasma half-life was 46.1 hours1. These are animal measurements from the original discovery work, not human data. Frequently asked questions
Why is semaglutide's mass so much larger than its peptide chain suggests?
The certificate's theoretical mass, 4113.6 Da, includes the side chain on lysine 26 as well as the 31 residues. A mass calculated from the amino acid sequence alone leaves out the diacid, the two spacers and the γ-glutamic acid, so it will not match the LC-MS result.
What does a 15 mg vial hold in moles?
Roughly 3.6 µmol: 15 mg over a molecular mass of 4113.6 Da. The powder also carries some residual water, reported as a percentage on the certificate, so the peptide itself comes in a little under that figure.
