This Epitalon nasal spray supplies the synthetic tetrapeptide AEDG, sequence Ala-Glu-Asp-Gly, molecular formula C14H22N4O9, molecular weight 390.35 Da, CAS 307297-39-8. It was designed at the St Petersburg Institute of Bioregulation and Gerontology by Vladimir Khavinson’s group as a simplified four-residue analog of epithalamin, an extract of bovine pineal gland. Almost everything written about this compound online repeats claims from that one laboratory. Something changed in 2025, and that is what this page leads with. The 2025 independent replication, and what it actually found

Until recently the honest summary of Epitalon was that the telomerase findings came from a single group and had not been reproduced by anyone unaffiliated with it. That is no longer quite true. A team at Brunel University London published Epitalon increases telomere length in human cell lines through telomerase upregulation or ALT activity in Biogerontology in 2025, testing four human cell types: two breast cancer lines (21NT and BT474), normal mammary epithelial cells (HMEC) and normal fibroblasts (IBR.3), with dose-response across multiple concentrations and durations.

The result splits by cell type, which is the interesting part. In the normal cells, the peptide raised hTERT transcript, increased telomerase activity and lengthened telomeres, which is the canonical telomerase route and matches what the original group reported. In the two telomerase-positive cancer lines, telomere lengthening happened instead through activation of ALT, the alternative lengthening of telomeres pathway. ALT was not activated in the normal cells.

I am flagging the cancer-line result rather than quietly leaving it out. A compound that switches on a telomere-maintenance pathway in transformed cells is a meaningful in-vitro observation, and anyone designing work with this peptide should know the finding exists. It is a cell-culture result and it does not translate directly to any claim about organisms, in either direction.

Read the correction notice too

That paper carries a correction. It was issued on 15 November 2025 and is indexed as PMID 41240216, with the full notice at PMC12619744. The wrong figures appeared as Figures 1, 2 and 3 in the original article, and the correction supplies the correct versions. The stated conclusions about telomere length were not withdrawn.

A figure error is not fabrication, and this is a normal part of how publishing self-corrects. It is still the version of the record you should cite, so pull the corrected figures rather than the originals if you are building on this work. Citing an uncorrected figure set is the kind of thing a reviewer notices.

What role do peptides play in experiments focusing on cellular senescence and repair?

Short peptides are used in senescence work mainly as probes for the pathways that set replicative lifespan, and Epitalon sits in the telomere-maintenance corner of that field. The standard readouts are telomere length by qPCR or by immunofluorescence, hTERT transcript by qPCR, direct telomerase activity assays such as TRAP, and senescence markers including senescence-associated beta-galactosidase staining and the p16 and p21 axis.

The reason this design keeps recurring is that telomere shortening is one of the few mechanisms of replicative senescence that can be measured directly in a dish over serial passage. Population doubling counts, passage number and donor age all move these endpoints on their own, so a senescence experiment without tight passage control produces numbers that cannot be compared with anyone else’s. That is a general weakness in Epitalon in vitro research, not a problem specific to this compound.

Where the wider evidence base actually stands

The originating claim is Khavinson and colleagues (2003), reporting that the peptide induced telomerase activity and telomere elongation in human somatic cells. Around it sits a large body of work from the same institute covering pineal function, melatonin rhythm and lifespan endpoints in rodents.

Two limitations are worth stating plainly. The concentration of the evidence within one group has been the central criticism of this literature for years, and the 2025 Brunel work is the first substantial independent replication of the cell-culture telomerase finding, not a general confirmation of everything else attributed to the compound. Separately, there are no independently replicated controlled human trials showing that systemic administration lengthens telomeres in people. Cell-culture evidence and organism-level claims are different things, and most pages selling this peptide blur them.

What that leaves is a well-defined small peptide with a reproducible in-vitro effect on telomere biology in at least one independent laboratory, and a much weaker case for anything beyond that. For a research program, the reproducible in-vitro effect is the usable part.

How telomerase and telomere length are actually measured

Anyone evaluating claims about this peptide needs to know that “telomere length went up” can mean several different measurements that do not agree with each other in absolute terms. Four methods dominate.

Telomerase activity is measured by the telomeric repeat amplification protocol, the TRAP assay, which detects the enzyme extending a synthetic substrate and amplifies the product. It reports enzyme activity in a lysate, not telomere length. Separately, hTERT transcript by qPCR reports whether the catalytic subunit is being transcribed, which is upstream of activity and can move independently of it.

Telomere length itself is measured either by quantitative PCR as a telomere-to-single-copy-gene ratio, which is relative and cheap and by far the most common, or by Southern blot of terminal restriction fragments, which gives absolute length in kilobases and is the older reference method, or by quantitative FISH, which gives per-cell and per-chromosome resolution. The qPCR ratio is sensitive to DNA quality and to plate-to-plate variation, so results from it travel poorly between labs.

The 2025 Brunel study used qPCR and immunofluorescence for length alongside hTERT qPCR and telomerase activity, and separately looked for ALT markers, which is the right combination: an upstream transcript readout, a direct enzyme readout, a length readout, and a check for the alternative pathway. If you are designing Epitalon telomerase research, that four-part panel is the standard to match, because a single length measurement on its own cannot tell you which mechanism produced the change, or whether anything changed at all.

Epitalon storage and handling

AEDG is a four-residue peptide with no cysteine and no methionine, so it avoids the disulfide and oxidation problems that dominate handling for larger peptides. That makes it comparatively forgiving. [CONFIRM: whether this SKU ships pre-filled in solution or as lyophilized material with an applicator.]

Two things still matter. Three of the four residues carry charged or polar side chains, with two acidic residues, so pH and ionic strength affect solubility and behaviour in solution more than they would for a neutral peptide. Check that your assay buffer holds pH where you think it does. And short peptides are cleared quickly by peptidases, so in serum-containing medium over long incubations you are working with a declining concentration rather than a fixed one, which is worth accounting for in any experiment that runs for days rather than hours.

The routine points apply as always: keep it cold, sealed and out of light, work from single-use aliquots rather than repeatedly freezing and thawing one container, and use low-binding plasticware at low working concentrations.
